How long does modafinil actually last? Real-world vs clinical data

Clinical trials cite a half-life of 12–15 hours. But what does that actually mean in terms of when you feel it and when you do not?

The timeline for 200 mg taken at 8 am:

  • 8:00 am — Take dose
  • 8:45–9:15 am — Onset (noticeable wakefulness and mild alerting)
  • 10:00 am — Peak begins
  • 10:00 am–4:00 pm — Full effect window, stable and strong
  • 4:00–6:00 pm — Plateau/taper begins, still clearly effective
  • 6:00–9:00 pm — Clearly declining but still more alert than baseline
  • 9:00 pm onward — Most people feel it is mostly worn off subjectively
  • Midnight — Pharmacologically ~50–60 mg equivalent still active (half-life math)

The disconnect: you may feel like modafinil has worn off by 9–10 pm, but pharmacologically enough is still circulating to suppress sleep quality even when you feel sleepy.

The disconnect between subjective “worn off” and pharmacological activity is the most important thing to understand about modafinil and sleep. This explains why people say “I slept fine” but their sleep tracker shows fragmented or shallow sleep.

  2. The midnight pharmacological activity point is why I set a 7 am hard cutoff for dosing. I need to be asleep by 10–10:30 pm.

  3. Does armodafinil have a longer or shorter effective duration? I know it has a different pharmacokinetic profile but have not found clear real-world comparisons.

    1. Armodafinil generally has a longer late-phase duration. The R-enantiomer (armodafinil) is cleared more slowly than the S-enantiomer (which is the other half of modafinil). So 150 mg armodafinil often feels like it lasts 14–16 hours vs 12 for modafinil.

Would taking a sleep aid (melatonin, magnesium) override the sleep-suppressing effect of residual modafinil?

    1. Melatonin helps with sleep onset timing. Magnesium glycinate helps with relaxation. Neither fully counteracts modafinil’s slow-wave sleep suppression, but both can meaningfully improve overall sleep quality. Worth using both if dosing later than ideal.

  5. Correct. Modafinil suppresses slow-wave sleep (deep sleep) even at sub-perceptual concentrations. You may sleep for 7 hours but not get the full restorative benefit.
